白细胞介素-17在肾组织的表达与糖尿病肾病关系探讨

摘    要:目的:观察白细胞介素-17(IL-17)在糖尿病大鼠不同时期血清、尿液中水平及肾脏表达,探讨其与糖尿病肾病发病相关性。方法:64只SD大鼠随机分为对照组及糖尿病组,以链脲佐菌素(50mg/kg)腹腔注射制备糖尿病大鼠模型。于8周、12周末采用ELISA方法检测血清、尿液中IL-17水平;测尿白蛋白排泄率(UAER)、血糖、血肌酐(Scr)、C反应蛋白(CRP);免疫组织化学染色观察肾组织中IL-17、转化生长因子-β1(TGF-β1)表达。结果:与对照组比较,糖尿病组血清、尿液中IL-17水平8周开始升高,肾组织中IL-17、TGF-β1表达增加,12周升高更明显;相关分析显示肾组织中IL-17表达与TGF-β1表达及尿液中IL-17水平、UAER、血CRP均呈正相关。结论:IL-17在糖尿病大鼠肾小管间质中表达随病程延长而增加,可能参与糖尿病肾病肾小管间质病变的发生发展。

Relationship Between the Expression of Interleukin-17 in the Kidney of Diabetic Rats and Diabetic Nephropathy

Abstract:Objective:To know the plasma and urine level and the expression of interleukin-17 in the kidney of diabetic rats,and their correlation with the renal injury.Methods:64 female Sprague-Dawley rats were randomly divided into two groups:control group and diabetes group.Rat diabetic nephropathy was induced by intraperitoneal injection of 50 mg/kg streptozoticin(STZ).In the 8th,12th week,serum and urine IL-17 were measured with sandwich ELISA.Meantime,CRP,the urinary albumin excretive rate(UAER),serum sugar,serum creatine (Scr) were measured.The expression of IL-17,TGF-β1 in the kidney were assessed by immunohistochemistry.Results:In comparison with control group,serum and urine IL-17 level,and the expression of IL-17 and TGF-β1 in rena1 tissue increased in diabetic rats;the elevated level of IL-17 in diabetic rena1 tissue were strongly correlated with expression of TGF-β1 in rena1 tissue,urine IL-17 level,UAER,CRP level.Conclusion:These results suggest that IL-17 may play an important role in the mediation of progressive renal injuries in the renal interstitial fibrosis of diabetic nephropathy,and its expression in kidney may indicate the disease activity and progressive renal injuries.
