大蓟、小蓟中黄酮类成分含量测定及色谱分析

摘    要:目的 对大蓟、小蓟中黄酮类成份的含量进行测定 ,并对其中的成分进行分离和鉴别。方法 采用紫外分光光度法对大蓟、小蓟中黄酮类成分进行含量测定 ,并采用反相高效色谱法对其中的成分进行分离和鉴别。结果 采用紫外分光光度法测定大蓟、小蓟中总黄酮含量时 ,芦丁对照品在 8 0 4 0~ 312 6 0 0 μg·ml-1范围内呈良好的线性关系 (r =0 9999) ,平均回收率分别为 10 0 5 3%、99 87% ;采用反相高效液相色谱法 ,各组分峰分离较好 ,特征峰明显。结论 分光光度法方法简便、准确、重现性好 ,结合反相高效液相色谱法可以对大蓟、小蓟这两味药材进行质量控制

Abstract:Objective To determine the concent of total flavones in Crisium japanisum and Crisium segetum,and to separate and distinguish some contents of flavones.Methods The concent of flavones in Crisium japanisum and Crisium segetum was determined by spectrophotometry.The content of flavones in Crisium japanisum and Crisium segetum was separated and distinguished by RP-HPLC.Results The linear range varied from 8.040~312.600μg·ml-1(r=0.99993),the average recovery was 100.53% and 99.87% respectively for Crisium japanisum and Crisium segetum,assayed by the spectrophotometry with the standard sample of Rutin.And the resolution was very high by RP-HPLC,and the specific peak was obvious.Conclusion Spectrophotometry method is simple,accurate and with high reproducibility.So we used it with RP-HPLC to control the quality of Crisium japanisum and Crisium segetum.
