摘    要:目的探讨有家族史食管癌患者血清蛋白的表达差异及其与食管癌遗传易患性之间的关系。方法应用表面增强激光解析电离飞行时间质谱技术结合铜离子蛋白芯片获得20例有家族史和26例无家族史食管癌患者的血清蛋白质谱。将获得的蛋白质谱采用Ciphergen公司的Biomarker Wizard和Biomarker Pattern软件分析。结果共找到10个表达差异蛋白,且此10个蛋白的含量在家族史阳性组均显著上调。其中,以质荷比为5344.77和7571.19建立的分类树模型能有效区别家族史阳性者和阴性者。在学习模式下分组正确率达到100%,在测试模式下分组正确率分别为100%(20/20)和96.2%(25/26)。结论质荷比为5344.77和7571.19的两个蛋白可能是影响食管癌遗传易患性的重要血清相关蛋白。

Screening mark proteins in serum from the esophageal cancer patients with family history by using SELDI-TOF-MS

Abstract:Objective To study the difference of proteomic spectra in serum from the esophageal cancer (EC) patients with or without family history, and to explore the relation of these changes with EC hereditary susceptibility. Methods Proteomic spectra of 20 serum samples from esophageal cancer with family history and 26 from EC without family history were generated by 1MAC-Cu pruteinchip array and SELDI-TOF-MS (surface enhanced laser desorption/ionization- time of flight). The discriminatory profiling was analyzed by Biomarker Wizard software and Biomarker Pattern software. Results Total 10 distinct proteins were found, and they were all high-expressed in the team with family history. Two proteins (M/Z 5 344.77 , M/Z 7 571.19) were choosed to make up a classification tree mode which can discriminate the two teams correctly. In learning mode the correct ratio was 100% and in test mode the ratios were respectively 100%(20/20) and 96.2%(25/26). Conclusion The two proteins (M/Z 5 344.77, M/Z 7 571.19) maybe play an important role in the EC family hereditary susceptivity mode.
