阿司匹林不同剂量和给药方法的药动学和药效学研究

摘    要:目的研究大鼠于不同时间间隔,服用不同剂量阿司匹林后,鼠血浆中水杨酸浓度及血小板聚集率的变化。方法35只SD大鼠随机分为对照组及实验组1(10 mg.kg-1.d-1,隔天一次)、实验组2(10 mg.kg-1.d-1,每天一次)、实验组3(30mg.kg-1.d-1,隔天一次)、实验组4(30mg.kg-1.d-1,每天一次),连续灌服阿司匹林7次后取血,观察各组大鼠血浆中水杨酸浓度及血小板聚集率的改变。结果小剂量阿司匹林每天一次用药与大剂量阿司匹林隔天一次用药后血浆中水杨酸浓度及二磷酸腺苷和花生四烯酸诱导下血小板聚集率比较差异无统计学意义(P>0.05),而与大剂量阿司匹林每天一次用药后相比均有显著差异(P<0.01)。结论小剂量阿司匹林每天一次与大剂量阿司匹林隔天一次给药能起到同样的抗血小板聚集作用。

Abstract:Objective To study the plasma concentrations of salicylic acid and changes of platelet aggregation rate(PAG) in SD rats after taking different doses of aspirin in different time.Methods 35 SD rats were randomly divided into the control group and different aspirin groups(group 1 with 10mg·kg-1·d-1 qod,group 2 with 10 mg·kg-1·d-1 qd,group 3 with 30mg·kg-1·d-1 qod,and group 4 with 30mg·kg-1·d-1,qd).After giving aspirin 7 times serially,Blood was sampled to test the concentrations of salicylic acid and platelet aggregation rate(PAG).Results There was no significiant difference in the plasma concentrations of salicylic acid and the platelet aggregation rate(PAG) induced by arachidonic acid and adenosine diphosphate between aspirin group 2 and aspirin group 3(P>0.05),whereas the difference was significiant between the former two groups and group 4(P<0.01).Conclusion Both aspirin 10mg·kg-1·d-1 qd and aspirin 30mg·kg-1·d-1 qod could inhibit platelet aggregation equally.
