Duffy血型的基因分型

摘    要:目的 建立Duffy血型基因分型方法。方法 根据Duffy血型cDNA结构,合成分型引物,建立聚合酶链反应-序列特异性引物(PCR-SSP)分型方法,对102例浙江省汉族无血缘关系的献血者进行基因分型。结果 PCR-SSP分型方法与血清学分型方法结果完全一致,102例浙江省汉族人,Fy^a纯合子91例(89.22%),Fy^aFy^b杂合子10例(9.80%),Fy^b纯合子1例(0.98%)。Fy^b基因频率0.9412,Fy^b基因频率0.0588。结论 PCR-SSP对Duffy血型基因分型,具有方法可靠、稳定,试剂来源广泛等优点,适合于临床常规应用,浙江省汉族Duffy血型基因以Fy^a为主。

Abstract:Objective To establish the genotyping method of the Duffy blood group.Methods According to the sequence of cDNA coding for FY glycoprotein,the Fy a/Fy b primers were synthesized,and the genotyping method with PCR SSP was established, The Duffy blood group of 102 unrelated donors in Zhejiang have been investigated by this method.Results Complete correlation between Fy phenotypes and genotypes was obtained in all samples studied. In all of 102 Zhejiang Han populations, 91 were Fy a/Fy a homozygotes(89 2%), 10 were Fy a/Fy b heterozygotes(8 8%),and 1 was Fy b/Fy b homozygote(0 98%).The gene frequency of Fy a was 0 9412,compared with that 0 0588 of Fy b.Conclution The genotyping method of Duffy blood group with PCR SSP was simple and rapid, and suitable for clinical application. The Fy a gene was the major gene of Duffy blood group in Zhejiang Han population.
