摘    要:目的 观察、探讨自体外周血单个核细胞(PBMC)对原代肝癌细胞生长的影响及芦荟多糖AP11的干预作用.方法 以手术切除肝癌组织为标本,胰酶短暂消化加吹打、单细胞悬液培养法原代培养肝癌细胞.通过倒置显微镜观察其形态学特征.于分离培养第2天,加芦荟多糖AP11、芦荟多糖AP11干预及未干预的PBMC于癌细胞中,倒置显微镜观察其对癌细胞形态的影响,MTT法观察其对癌细胞生长的影响.结果 采用胰酶短暂消化加吹打分离、单细胞悬液原代培养的方法,可成功获取原代肝癌细胞;与空白对照组相比,芦荟多糖组癌细胞在形态上无明显差别,MTT实验结果表明其对癌细胞的生长无明显影响;与空白对照组相比,芦荟多糖AP11干预与未干预PBMC组癌细胞形态均有较大变化,MTT实验结果表明,其对癌细胞的生长有显著抑制作用.结论 芦荟多糖AP11无直接体外抗肿瘤活性,其抗肿瘤活性可能是通过与PBMC的相互作用,通过调节机体的免疫系统而达到的.

Abstract:Objective To observe the effect of PBMC on homebody primary hepatoma cells and the intervention of Aloe vera polysaccharide.Methods With hepatoma tissue as sample,primary culture hepatoma cells were isolated by using the method of pancreatic enzyme transient digestion and simultaneous blowing,and its morphology was observed by inverted microscope.Then AP,PBMC and PBMC dealt with AP were added to primary culture hepatoma cells after two-day cultivation,and the effect on carcinoma cell morphology was observed...
