摘    要:在心脏中细胞内游离Ca2 是一种关键的第二信使,Ca2 稳态的改变会显著影响许多心脏功能包括收缩、舒张和心脏发育。钙离子在内质网中主要与钙网织蛋白(calreticulin,CRT)结合,钙网织蛋白水平的改变显著影响细胞内Ca2 稳态,其在胚胎发育过程中丰度的变化以及crt基因敲除可损伤心脏发育而导致胚胎死亡揭示了CRT在心脏发育中所起的重要作用。crt缺陷揭示了Ca2 依赖的控制点是心肌纤维形成过程中早期事件的关键。

Abstract:Intracellular free Ca2 is a critical second messenger in the heart.Alterations in intracellular Ca2 homeostasis have profound effects on cardiac functions including contraction-relaxation and cardiac development.Within the ER,Ca2 is mainly bound to calreticulin(CRT).The alterations of its abundance in the heart during the embryogenesis and embryonical death due to impaired cardiac development caused by crt knockout revealed the importance of CRT in the cardiac development.We are led to conclude that the absence of CRT interferes with myofibril formation,suggesting that a Ca2 -dependent checkpoint is critical for early events during cardiac myofibrillogenesis.
