葛根素对缺氧复氧心脏微血管内皮细胞的保护作用

摘    要:目的:探讨葛根素在心脏微血管内皮细胞(cardiac microvascular endothelial cell,CMEC)缺氧复氧损伤中的作用。方法:将CMEC随机分为空白组、模型组、葛根素低、中、高剂量(25,50,100 mg·L-1)共3组。四甲基偶氮唑(methylthiazolyl tetrazolium,MTT)法检测细胞活力,流式细胞仪检测细胞凋亡,比色法检测乳酸脱氢酶(lactate dehydrogenase,LDH)活性、丙二醛(malondialdehyde,MDA)含量及超氧化物歧化酶(superoxide dismutase,SOD)活性,Western blot法检测Caspase-3和Bcl-2蛋白表达。结果:葛根素预处理后,缺氧复氧CMEC细胞活力增强,细胞凋亡率、LDH活性及MDA含量降低,SOD活性增强,Caspase-3蛋白表达减少,Bcl-2蛋白表达增加。结论:葛根素可拮抗缺氧复氧所致的CMEC损伤,可能与其抗氧化及抗凋亡活性相关。

Abstract:OBJECTIVE To discuss effects of puerarin on hypoxia/reoxygenation of cardiac microvascular endothelial cells (CMEC).METHODS CMECs were randomly divided into blank group, model group and low, medium and high puerarin dose (25, 50, 100 mg·L-1) groups. Cell viability was determined with methylthiazolyl tetrazolium (MTT) method, cell apoptosis was detected by flow cytometry, lactate dehydrogenase (LDH) activity, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were determined by colorimetry, and protein expressions of Caspase-3 and Bcl-2 were detected by Western blot method.RESULTS After precondition of puerarin, cell viability of CMEC dealt with hypoxia/reoxygenation enhanced, cell apoptosis rate, LDA activity and MDA content decreased, SOD activity increased, Caspase-3 protein expression reduced and Bcl-2 protein expression increased.CONCLUSION Puerarin can antagonize CMEC injury induced by hypoxia/reoxygenation, which may be related to its antioxidant and apoptotic activity.
