包埋前染色免疫酶电镜技术及其应用

摘    要:以辛德比斯病毒(SIN病毒)感染地鼠肾传代细胞(BHK-13)为模型,建立了包埋前染色免疫酶电镜技术。应用于绿猴肾细胞(Vero E-6)上观察到从肾综合征出血热(HFRS)病人血中直接分离出的HFRS病毒颗粒;用于新疆精河株斑点热立克次体结构的研究,观察到在常规超薄切片电镜中难以见到的立克次体的表面结构;还对大白鼠腘窝淋巴结浆细胞内产生的特异抗体作了鉴定和定位观察。


THE TECHNIQUE OF PRE-EMBEDDING STAINING OF IMMUNOENZYMATIC ELECTRON MICROSCOPY AND IT' S APPLICATIONS
Abstract:The pre-embedding staining method of immunoenzymatic electron microscopy was established on BHK-13 cells infected with Sindbis virus. We have observed HFRS virus particles isolated in Vero E-6 cells from a patient with HFES by this method. Moreover, we have also successfully applied this mehtod to the observation of surface structure of Jing-He strain of spotted fever rickettsiae in the yolk sac section which might not be detected with ordinary electron microscopy and the localization of specific antibody of plasma cells in rat popliteal lymph nodes after immunization with horseradish peroxidase.
