雄激素及其选择性受体调节剂对PC12细胞Seladin-1表达的影响

目的 观察雄激素和AR调节剂干预对Seladin-1表达的调控作用,以初步探讨其作用机制.方法 将培养的PC12细胞分为两组：雄激素、AR激动剂组和雄激素、AR阻断剂组,两组均设空白对照组.采用Western-blot和Real-time PCR方法对PC12细胞Seladin-1的表达进行检测.结果 AR激动剂组处理后,PC12细胞Seladin-1的蛋白质与Mrna的表达水平与正常对照组比较均明显提高,差异具有统计学意义（P〈0.05,P〈0.001）.AR阻断剂组处理后,PC12细胞Seladin-1的蛋白质与mRNA的表达水平与正常对照组比较均明显降低,差异具有统计学意义（P〈0.05）.结论 雄激素和AR参与了PC12细胞Seladin-1表达的调控,雄激素的神经保护效应可能是与通过和AR结合形成激素-受体复合物,并上调seladin-1的表达有关.

The effects of androgen and selective androgen receptor modulators on the expression of Seladin-1 in PC12 cells

Objective To investigate the effects and the mechanisms of androgen and androgen receptor modulators on the expression of Seladin- 1. Methods The expression of Seladin- 1 in PC12 cells, which was treated with androgen, androgen receptor agonist and androgen receptor antagonist, was detected by means of Western blot and Real-time PCR. Results The expression of Seladin- 1 in PC12 cells was significantly increased by androgen receptor agonist both in protein levels （P 〈 0.05） and mRNA levels （P 〈 0.00l）, while it was decreased by androgen receptor antagonist with significant difference （P〈 0.05）. Conclusions Androgen and androgen receptor agonist were involved in the modulation of the expression of Seladin--1 in PC12 cells. The neuroprocetive effect of androgen was associated with the up-regulation of seladin- 1 which was mediated by the combination of androgen with androgen receptor.
