摘    要:①目的探讨建立轮状病毒的分离与培养方法。②方法粪便标本经ELISA检测,离心后接种于MA104细胞株,观察病毒致细胞病变效应。提取病毒RNA.琼脂糖电泳检测。③结果经离心、双抗处理后,轮状病毒可在MA104细胞中克隆增殖.并引起细胞病变(CPE)。④结论MA104作为容纳细胞,可用于轮状病毒的大量扩增。

Abstract:Objective To establish a method to isolate and incubate rotavirus. Method Specimen were detected by ELISA and inoculated to MA104, then the CPE caused by virus was observed. Total RNA of virus was extracted and agar sugar electrophoresis was performed. Result After treated by centrifugation and bio-antibiotics, rotavirus was isolated from stool and proliferated in MA104 cells and caused CPE as a result. Conclusion As a kind of adoptive cell, MA104 can be used for rotavirus abundance proliferation.
