HBeAg阳性和HBeAg阴性患者树突状细胞的功能

目的比较HBeAg阳性和HBeAg阴性患者外周血DC的功能差异,为两种患者经DC免疫治疗的预后提供参考。方法用rhGM-CSF和IL-4体外诱导乙肝患者外周血中的单核细胞为树突状细胞,流式细胞仪检测DC表型,MTT法检测DC刺激同种异体混合淋巴细胞反应强度,ELISA法检测DC分泌IL-6和IL-12的水平。结果细胞成熟度、对同种异体淋巴细胞的刺激强度、IL-12的分泌水平,HBeAg阳性组和HBeAg阴性组均显著低于健康对照组（P〈0.01）,而IL-6的水平则显著高于健康对照组（P〈0.01）。HBeAg阳性组的CD83水平、对同种异体T淋巴细胞的刺激指数和IL-12的分泌水平均高于HBeAg阴性组（P〈0.05）,其他指标的差异无统计学意义。结论与健康人相比,乙肝患者外周血DC存在功能上的缺陷;而HBeAg阳性患者外周血DC的功能要强于HBeAg阴性患者,接受DC免疫治疗预后效果可能更好。

Function of Dendritic cells of HBeAg positive and negative patients

Objective To compare the function of Dendritic cells of HBeAg positive and HBeAg negative patients,so as to predict the effects of DC immunotherapy.Methods DCs were enriched from peripheral blood monomuclear cells by density grade centrifugation and induced by rhGM-CSF and IL-4.The phenotypes were measured by flow cytometry.The antigen presenting activity was evaluated by the stimulation index on homogeneity T lymphocyte.The cytokines were detected by enzyme linked immunosorbent assay.Results The express levels of CD83、CD80 and CD86 on DCs derived from the patients were significantly lower than those from the normal（P0.01）,so did the stimulate index and the IL-12.comparing with the DCs from HBeAg negative patients,DCs from HBeAg positive patients had more activity to stimulate the homogeneity T lymphocyte and to produce IL-12（P0.05）.Conclusion The DCs from CHB patients were defective.The DCs from HBeAg positive patients were more active than those from HBeAg negative patients,which indicated that the HBeAg positive patients may get a better curative effect if they accepted DC immunotherapy.
