藏花素调控deltaNP63基因抑制膀胱肿瘤细胞生长的研究

摘    要:目的探讨藏花素对膀胱肿瘤细胞5637的生长抑制作用及调控机制。方法用3mmol/L藏花素处理膀胱肿瘤细胞5637细胞株,采用反转录PCR(RT-PCR)检测细胞deltaN P63基因表达的变化,光镜下检测细胞形态改变,流式细胞术检测细胞周期改变,四甲基偶氮唑盐法(MTT)检测细胞增殖改变。结果在一定浓度的藏花素作用下,可见5637细胞生长缓慢,deltaN—P63基因表达下降,G0/G1期细胞数量增加,G2/M期细胞数量减少,MTT检测显示细胞增殖受到抑制。结论藏花素可以抑制膀胱肿瘤细胞增殖和生长,其机制可能与通过下调deltaN P63基因的表达有关。

Crocin inhibits bladder cell growth through regulation of deltaN P63 gene

Abstract:Objective To detect the inhibition effects and possible mechanism of crocin in bladder cancer 5637 cell line. Methods 5637 cells were treated with 3 mmol/L crocin,and the change of deltaN P63 gene was detected using reverse transcription-polymerase chain reaction (RT-PCR) ,the cell form was observed by microscope, cell cycle was detected by flow cytometry, and cell pro- liferation was detected by MTT. Results In the optimal concentration of crocin,the growth of 5637 cells became slowly than nor- mal. The expression of deltaN P63 gene decreased, the cell number of G0/G1 was increased but the cell number of Gz/M decreased. MTT result showed that the cell proliferation was inhibited. Conclusion Bladder cancer cell growth and proliferation could be in- hibited by crocin,and its possible mechanism is the down regulation of deltaN P63 gene.
