缺氧适应与缺血低氧对c-fos和c-jun表达的影响

摘    要:目的:探讨缺氧适应对缺血低氧脑c-fos,c-jun基图表达的影响。方法:昆明纯种雄性小白鼠36只随机分组:(1)对照组;(2)缺血低氧组(IH);(3)缺氧适应与缺血低氧组(HA—IH)。采用免疫组化技术检测c-fos,c-jun基因表达。结果:IH组C—fos和c-jun阳性细胞在30min均呈低密度分布,c—fos在1h表达高峰,12h基本消失,而c-jun基因的表达高峰在3h;与IH组比较,HA—IH组c—fos阳性细胞数减少,c-jun表达增加。结论:缺血低氧可诱发中枢神经系统c-fos、c-jun基因表达,且有时间依赖性;缺氧适应可抑制缺血低氧脑c-fos基因的表达,增强c-jun基因的表达。

Abstract:Objective: To explore the effect of hypoxic adaptation on c-fos and c-jun gene expressions in cerebral hypoxia-ischemia.Methods: Thirty-six mice were randomly divided into 3 groups: control group,hypoxia-ischemia(IH) group,and hypoxic adaptation plus hypoxia-ischemia(HA-IH) group.The expressions of c-fos and c-jun genes in brain were determined with immunohistochemical S-P method.Results: No expression of c-fos and c-jun genes was found in the control group.In IH group,the cells positive for c-fos and c-jun scattered 30 minutes after hypoxia-ischemia;the expression of c-fos reached the peak 1 hour after hypoxia-ischemia,declined 3 hours later,and disappeared 12 hours later;the expression of c-jun reached the peak 3 hours after hypoxia-ischmia.Compared with IH group,the expression of c-fos increased and the expression of c-jun decreased in HA-IH group.Conclusion: Hypoxia-ischemia could induce c-fos and c-jun gene expressions in central nervous system in a time-dependent manner.Hypoxic adaptation may inhibit(c-fos) gene expression and increase c-jun gene expression.
