磷脂酶A2与高血压大动脉血管重构关系初步研究

摘    要:目的探讨磷酸化胞浆型磷脂酶A2(cPLA2)及其上游激酶MEK与高血压大动脉血管平滑肌细胞增殖/凋亡失衡的关系,进一步探讨高血压状态下大动脉血管重构的可能机制。方法将4、16、24周龄组雄性自发性高血压大鼠(SHR)作为实验组,同周龄雄性Wistar大鼠(MKY)作为对照组,各组5只大鼠。采用免疫组化和Western blot对比分析各组动物主动脉平滑肌细胞中cPLA2以及MEK的表达。结果 (1)SHR从8周龄开始血压升高,SHR16、SHR24与同周龄WKY相比差异有统计学意义(P<0.05);(2)各周龄WKY心脏/体质量比值差异无统计学意义(P>0.05),相同周龄SHR心脏/体质量比值较高并随周龄增加心脏/体质量比值呈增高趋势,SHR心脏/体质量比值均高于同周龄对照组;(3)磷酸化cPLA2在SHR4表达率分别为11.71%、46.00%、51.59%,而同周龄WKY表达率分别为29.10%、36.00%、67.84%,随大鼠周龄增加,磷酸化cPLA2表达率逐渐增加(P<0.05);(4)随大鼠周龄增加,磷酸化MEK表达率增加(P<0.05)。结论 MEK的激活与cPLA2活性减弱可能共同参与了高血...
