PCNA在胆管癌中的表达及意义

目的探讨PCNA其在胆管癌发生发展中的作用及机制,为胆管癌诊断和治疗寻找新的靶点。方法分别采用免疫组化SP法和RT-PCR法检测PCNA蛋白和mRNA在胆管癌组织、癌旁组织及正常组织中的表达,结合患者的年龄、性别以及肿瘤大小、部位、分化程度、淋巴结转移等临床病理因素进行综合分析。结果胆管癌组织、正常胆管组织中PCNA蛋白表达的阳性率分别为71.19%和25.00%,胆管癌组织的阳性率亦明显高于正常组织(P<0.05);胆管癌组织的PCNAmRNA表达高于正常组织(P<0.05)。结论 PCNA在胆管癌中的表达明显高于正常的胆管组织,且PCNA的表达随着肿瘤恶性程度的升高(即肿瘤的病理学分化程度的降低)而升高,说明了PCNA是一个有价值的肿瘤标志。

The expression and significance of PCNA in cholangiocarcinoma

Objective To explore the action mechanism of PCAN in the pathogenesis and progress of cholangiocarcinoma,so as to search for new target point for the diagnosis and treatment of cholangiocarcinoma. Methods Immunohistochemical technique （SP） and RT-PCR were respectively used to detect the expression of PCNA protein and mRNA in cholan~ocarcinoma tissue and normal tissue. The expression levels of PCNA were analyzed, combined with the clinicopathological factors including patients ＇ age, sex, tunor＇ s size, position, differentiation degree and lymph node metastasis. Results The positive rate （71.19%） of protein expression of PCNA in cholangiocarcinoma tissue was significantly higher than that （25%） in normal bile duct tissue （ P 〈 O. 05）, and the expression levels of PCNA mRNA in carcinoma tissue were also significantly higher than those in normal bile duct tissue （ P 〈 0. 05 ）. Conclusion The expression levels of PCNA in carcinoma tissue are significantly higher than those in normal bile duct tissue,furthermore ,which increase with the malignant degree of tumor,so that PCNA is a valuable tumor marker.
