| 摘 要： | 目的筛选出与肝细胞有较好生物相容性的半透膜材料,以用于生物人工肝支持系统中空纤维型反应器的制备。方法选择聚醚砜(PES)、聚砜(PSF)、醋酸纤维素(CA)、聚偏二氟乙烯(PVDF)、聚丙烯(PP)、混合纤维素酯(CNA)等有代表性的6种半透膜材料进行肝细胞(L02细胞)培养,同时以聚苯乙烯培养板作为对照组,检测细胞LDH漏出量和DNA Ladder细胞凋亡评价膜材料的细胞毒性,观察各种膜材料上L02细胞的贴壁率、细胞活性、DNA总量等。结果各种膜材料未见明显细胞毒性;L02细胞在PES膜上的贴壁率显著高于CA、PVDF、PP、CNA膜(P<0.05),与对照组和PSF膜无显著差异;PES膜上贴壁细胞数明显较CA和CNA膜多;PES和PSF膜上细胞DNA总量显著高于CA、PP、CNA膜(P<0.05);在7天的培养时间内,PES膜上和对照组的细胞活性最高,显著高于CA、PVDF、PP、CNA膜(P<0.05)。结论在6种半透膜材料中,PES是肝细胞相容性最佳的半透膜材料,适宜用于生物反应器中空纤维的制备。
