| 摘 要： | 目的：研究Ｒｂ基因异常与急性髓系白血病发病的关系。方法：采用ＤＮＡ印迹杂交和ＰＣＲ技术。结果：在５／１８例原发急性髓系白血病中发现Ｒｂ基因结构异常（２８％），其中２例缺失５．４ｋｂ和出现新片段３．１ｋｂ和２．３ｋｂ。将其中１例用Ｒｂ基因外显子１８，１９，２１，２２，２７五对引物进行分析，未发现异常。结论：Ｒｂ基因异常可能与急性髓系白血病的始发和发展有关。

| Abstract: | Objective: To study the correlation between abnormality of Rb gene and initiation of AML. Methods: Southern blotting and PCR techniques were used in this study. Results: The structure of the Rb gene in 18 cases of acute myelogenous leukemia (AML) were studied. The structural abnormal alternations of Rb gene were found in 5 cases of AML (28%). Two novel fragments (3.1 kb and 2.3 kb) and deletion of 5.4 kb were observed in 2 cases. We used 5 pair of Rb gene primers of exon 18, 19, 21, 22, 27 and amplified Rb gene from one case of AML with abnormal Rb gene. No abnormality was found. Conclusion: Abnormalities of Rb gene might be associated with the initiation and progression of AML. |
