麦考酚酸产生菌原生质体诱变育种的研究

摘    要:对麦考酚酸产生菌(Penicillium brevicomactum)的原生质体制备、再生条件进行了研究,建立了原生质体诱变筛选体系。在此基础上,采用紫外线和亚硝酸复合诱变处理麦考酚酸产生菌的原生质体,经原生质体再生获得了A-S-8的菌株,该菌株比对照菌株生产能力提高28.2%,摇瓶和50L发酵实验的效价均可达到6000mg/L以上。

Abstract:To select high toplast preparation, regeneration and mycophenolic acid producing mutants from mutagenesis were carried out. The mutant Penicillium brevicompactum, the prostrain A-S-8 of Penicillium brevicompactum which possessed high yield of mycophenolic acid was obtained through combined induction with UV and HNO2. The mycophenolic acid yield of mutant was increased by 28.2% compared with the starting strain. The mycophenolic acid titre of strain A-S-8 reached above 6000mg/L both in shaking flask and 50-liter fermenter experiments.
