MicroRNA在大鼠心肌缺血再灌注损伤中表达谱的变化

摘    要:目的:分析微小RNA(mieroRNA)在大鼠心肌缺血再灌注损伤中表达谱的变化,为缺血再灌注损伤治疗中的microRNA干预策略提供可供选择的目标microRNA.方法:利用microRNA基因芯片技术检测大鼠正常心肌及缺血再灌注损伤心肌micmRNA表达谱的差异.实时定量PCR检测上述2组大鼠心肌组织中基因芯片结果差异性表达的microRNA,验证芯片结果的可信性.结果:与正常大鼠心肌组织相比,缺血再灌注心肌组织中表达上调的microRNA有10个,分别为mo-microRNA-21、mo-micmRNA-26b、mo-microRNA-499、mo-microRNA-126、mo-let-7e、mo-microRNA-23a、mo-microRNA-23b、mo-let-7a、mo-let-7d、mo-microRNA-26a;表达下调的microRNA有6个mo-microRNA-214、mo-microRNA-125b-5p、mo-microRNA-1、mo-microRNA-133a、mo-microRNA-133b、mo-microRNA-24.结论:缺血再灌注损伤中伴有microRNA表达谱的变化.

Abstract:Objective To investigate the changes of the expression profile of microRNAs in myocardium of rats with ischemia reperfusion injury. Methods The expression profiles of microRNA in normal myocardium and ischemia reperfusion injured myocardium of rats were detected by microRNA genetic chip, and were verified by real-time quantitative PCR. Results As compared with normal myocardium, 10 microRNAs: rno-microRNA-21, rno-microRNA-26b, rno-microRNA-499, rno-microRNA-126, rno-let-7e, rno-microRNA-23a, rno-microRNA-23b, rno-let-7a, rno-let-7d, and rno-microRNA-26a were up-regulated, and 6 microRNAs: rno-microRNA-214, rno-microRNA-125b-5p, rno-microRNA-1, rno-microRNA-133a, rno-microRNA-133b, and rno-microRNA-24 were down-regulated in the ischemia reperfusion injured myocardium. Conclusions Ischemia reperfusion injury is associated with the change of the expression profile of microRNAs.
