AngiostatinK(1-3)裸DNA治疗人脑胶质瘤的实验研究

探讨ＡｎｇｉｏｓｔａｔｉｎＫ（１－３）〖ＡＫ（１－３）〗裸ＤＮＡ治疗人脑胶质可行性和作用机制。用脂质体包埋法将ＡＫ（１－３）基因的真表达载体ｐｃＤＮＡ－ＳＡＫ（１－３）注入裸以下ＳＨＧ４４人脑胶质瘤的特点。ＡＫ（１－３）裸ＤＮＡ在荷瘤裸本内获得表达，裸鼠皮下胶质瘤血管生成能力显著下降，肿瘤生长受到抑制，抑瘤率达４３％。本研究证明，ＡＫ（１－３）裸ＤＮＡ能抑制人脑质瘤生长，为非病毒载体介导的抗血管生

ANEXPERIMENTAL RESEARCH OF HUMAN GLIOMAS THERAPY BY DIRECT TUMOR INJECTIONS OF EUKARYOTIC VECTOR ENCODING ANGIOSTATIN K (1-3) GENE

To probe the feasibility and mechanism of human glioma therapy bythe eukaryotic vector encoding angiostatin K(1 3) gene, simply named pcDNA SAK(1 3). The vector of pcDNA SAK(1 3) packaged by lipofectamine was injected into SHG44 human glioma of nude mice. The glioma growth was observed and thereduction in tumor size was calculated. The characteristic of gene therapy of gliomas was recognized according to the PCR of AK(1 3), immunohistochemistry, electron microscope, tumor necrosis and micrangium calculation. The pcDNA SAK(1 3) vectorwas successfully expressed in the tumor of nude mice. The tumor growth and angiogenesis of glioma cells in nude mice were greatly reduced. And the mice treated with pcDNA SAK(1 3) exhibited a reduction in tumor size of 43%. The study showed that human glioma angiogenesis and growth can be inhibited by the eukaryotic vector encoding angiostatin K(1 3) cDNA. These findings lays a foundation for the further application of nonviral delivery of antiangiogenic genes.
