摘    要:目的探讨核转录因子(NF)-κB p65的阻断剂SN50对裸鼠人胃癌移植瘤生长的影响。方法建立裸鼠胃癌移植瘤模型后将20只裸鼠随机分成对照组(n=5)和SN50实验组(n=15),用生理盐水处理对照组、SN50处理实验组,于第5 d、10 d、15 d分别取5只裸鼠测量肿瘤体积,计算抑瘤率。采用免疫组织化学染色观察凋亡指标Bcl-2、Bax和p53表达的变化。结果使用SN50处理肿瘤后5 d、10 d、15 d肿瘤生长抑制率逐渐增高;与对照组相比,凋亡指标Bcl-2的表达下调,p53和Bax的表达上调(均P〈0.05)。结论 NF-κB p65的阻断剂SN50可能通过下调Bcl-2的表达和上调p53、Bax的表达导致细胞凋亡,从而最终抑制移植瘤的生长。

Abstract:Objective To investigate the effects of NF-κB p65 inhibitor SN50 on the implanted tumor of human gastric cancer cell in nude mice.Methods Gastric cancer xenografts models were made in nude mice.Twenty nude mice were randomly divided into the control group(n=5) and SN50 treatment group(n=15,5 for 5 days,5 for 10 days,5 for 15 days) after the the models were made.The animals in the control group injected with physiological saline and in the experimental group with SN50.Five,ten and fifteen days after treatment,the bulk of the tumor and calculate inhibitory rate of tumor growth was measured.The expressions of Bcl-2,p53 and Bax were observed by immunohistochemical staining.Results Treating the tumor with SN50 for 5,10,15 days could inhibit carcinoma growth significantly.The expression of Bcl-2 was down-regulated and the expression of p53 and Bax were up-regulated.It was statistically significant compared to that of the control group(P0.05).Conclusion NF-κB p65 inhibitor SN50 could inhibit the growth of implanted tumor of human gastric cancer.
