Abstract:Objective:To investigate the effects of shRNA-ADAR1 on the proliferation of human K562 cell line.Methods:The expression of ADAR1 in patients with chronic myeloid leukemia(BP)and normal adults was detected by real-time fluorescence quantitative polymerase chain reaction and Western-blot.A stable K562 cell line(shADAR1)with knockdown ADAR1 was established by culturing human leukemia cell line K562 cells with lentivirus mediated siRNA.The expression of mRNA and protein of ADAR1 in K562 cells and sh ADAR1 cells was detected by qRT-PCR and Western-blot,and the proliferation of K562 cells was detected by MTT assay.Results:The expression of mRNA and protein level of ADAR1 in patients with chronic myeloid leukemia(BP)was significantly higher than that in normal adults(P<0.05).The expression of mRNA and protein in ADAR1 in shADAR1 cells was significantly lower than that in K562 cells(P<0.05).Down-regulation of ADAR1 could inhibit cell proliferation(P<0.05).Conclusion:The expression of ADAR1 is increased in patients with chronic myeloid leukemia(BP).Inhibition of ADAR1 expression by siRNA can significantly inhibit the proliferation of human leukemia cell line K562.ADAR1 gene may be a new target for the treatment of chronic myeloid leukemia.
