POD-SLN对人角质形成细胞凋亡的影响及其在细胞内的分布

摘    要:目的研究鬼臼毒素固体脂质纳米粒(POD-SLN)对人角质形成细胞凋亡的影响及在细胞内的分布。方法设鬼臼毒素(POD)组、POD-SLN组和对照组,分别处理人角质形成细胞,流式细胞术检测细胞凋亡和细胞周期,激光共聚焦显微镜(CLSM)观察POD-SLN在人角质形成细胞的俘获分布。结果 POD和POD-SLN均能够显著促进细胞凋亡,POD-SLN诱导细胞凋亡的效果优于POD,并且以早期凋亡为主(P〈0.01)。POD-SLN和POD均导致细胞周期阻滞,两组对细胞周期的影响差异无统计学意义(P〉0.05)。CLSM呈现红色荧光聚集在细胞核的倾向,并且POD-SLN较POD的荧光强度增高。结论 POD-SLN较POD对角质形成细胞具有更强的诱导凋亡的效应。

Abstract:Objective To study the effects of podophyllotoxin-loaded solid lipid nanoparticles(POD-SLN) on apoptosis of human keratinocytes and to observe the cellular uptake of POD-SLN in human keratinocytes in vitro.Methods The cell apoptosis and the cell cycle were examined with flow cytometry(FCM) technique.Cell uptake of free POD and POD delivered by solid lipid nanoparticles were confirmed by using confocal laser scanning microscopy(CLSM).Results Both POD-SLN and POD could significantly induce the apoptosis of human keratinocytes.POD-SLN inducing the cell apoptosis was better than that of POD,and especially in early apoptotic phase(P0.01).POD-SLN and POD blocked human keratinocytes in G2-M phase(P0.05).Red staining located in the cell nucleus of human keratinocytes,and the fluorescence intensity of POD-SLN was stronger than that of POD.Conclusion Compared with POD,POD-SLN had better effect on inducing the apoptosis of human keratinocytes.
