摘    要:目的 研究酚红在神经细胞培养中的弱雌激素样作用.方法 使用不同培养基(其一含酚红、另一无酚红)培养MN9D细胞,并分别于培养基中加入胶质源性神经营养因子(GDNF),然后加入相同浓度6-羟多巴胺(6-OHDA)进行损伤,最后四甲基偶氮唑蓝(MTT)比色法检测细胞的存活状况;提取不同培养基中的正常细胞,Western blot法检测雌激素α受体(ERα)在细胞内的移位.结果 培养基中没有酚红时的细胞存活率较有酚红时低,且当GDNF与酚红同时存在时,细胞的存活率比只有GDNF或酚红存在时高;酚红能够促使ERα从胞质转移至细胞膜.结论 酚红能够激活ERα,并具有一定的神经保护作用.

Abstract:Objective To investigate the weak estrogen - like effects of phenol red in neuronal culture. Methods Different media ( one with phenol red, the other without phenol red) were used for MN9D cell culture and glial - cell - line- derived neurotrophic factor (GDNF) was added in these two kinds of media respectively, followed by the addition of the same concentration of 6 -OHDA to induce injury to the neurons. Finally, MTT assay was used to detect the changes in cell survival. Normal MN9D cells were extracted from different media, and then Western blot was used to detect the shift of estrogen receptor a (ERa) inside the cells. Results The cell survival with phenol red in the medium was lower than that without phenol red in the medium. Cell survival with GDNF in the medium was higher than that with phenol red in the medium. With both GDNF and phenol red in medium, cell survival was the highest. With the action of phenol red, ERa was transferred from the cytoplasma to the cell membrane. Conclusion Phenol red can activate the ERa and has certain neuroprotective effect.
