白藜三醇对房颤所致猪心房组织中MMP-1、MMP-9、TIMP-1表达的影响

摘    要:目的 研究白藜三醇对心房纤颤(简称房颤)猪心房组织基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶组织抑制剂-1(TIMP-1)蛋白表达的影响,探讨白藜三醇抗心肌纤维化作用及可能机制.方法 18只小家猪通过起搏器植入法建立猪房颤模型,随机分为3组(每组6只):对照组、房颤组、白藜三醇干预组(2.5 mg·kg^-1·d^-1).苏木精-伊红染色、Masson染色法观察心房组织形态学和间质纤维化,Western blotting法检测心房组织中MMP-1、MMP-9、TIMP-1蛋白表达水平.结果 与对照组相比,房颤组MMP-1、MMP-9显著增加(P<0.05),而TIMP-1则显著降低(P<0.05);与房颤组相比,白藜三醇干预组MMP-1、MMP-9显著降低(P<0.05),而TIMP-1则显著增加(P<0.05).结论 白藜三醇可能通过调节心肌组织中MMP-1、MMP-9、TIMP-1的表达而发挥抗心肌纤维化的作用.

Abstract:Objective To investigate the effect of resveratrol on the expression of matrix metalloproteinase - 1 ( MMP - 1 ), matrix metalloproteinase - 9 ( MMP - 9) and tissue inhibitor of metalloproteinase - 1 ( TIMP - 1 ) in porcine with atrial fibrillation, and to explore the possible underlying mechanism of resveratrol against myocardial fibrosis. Methods Swine atrial fibrillation models were established by plantation of pacer in 18 piglets, which were randomly divided into 3 groups ( n = 6 each ) : control group, atrial fibrillation group and resveratrol - intervened group (2.5 mg·kg^-1· d^-1 ). The myocardial histomorphological changes and the degrees of atrial interstitial fibrosis were detected by hematoxylin - eosin staining and Masson staining. Western blotting was used to determine the expression levels of atrial MMP - 1, MMP - 9 and TIMP - 1. Results Compared with control group, MMP - 1 and MMP - 9 level significantly increased in atrial fibrillation group (P 〈 0.05 ), while TIMP - 1 level remarkably decreased ( P 〈 0.05 ). Compared with atrial fibrillation group, MMP - 1 and MMP - 9 expression decreased significantly in resveratrol - intervened group (P 〈 0.05), while TIMP - 1 level increased significantly ( P 〈 0.05). Conclusion, Resveratrol can inhibit the development of myocardial fibrosis, possibly via the downregulation of the expression of myocardial MMP - 1 and MMP - 9 and the upregulation of TLMP - 1 expression.
