丹皮酚对大鼠心肌缺血再灌注损伤保护中HMGBl表达的影响

摘    要:目的:探讨丹皮酚(paeonol,Pae)对大鼠心肌缺血再灌注损伤保护中HMGBl表达的影响。方法:清洁sD大鼠40只随机分为4组(/2=10):假手术组(Sham组)、缺血再灌注组(I/R组)、丹皮酚低剂量组(PaeLD组)及丹皮酚高剂量组(PaeHD组)。建立大鼠心肌缺血再灌注模型。TTC染色法检测大鼠梗死心肌体积,Tunnel法检测心肌缺血坏死区细胞凋亡;免疫印迹检测缺血坏死组织HMGBl的表达。结果:PaeLD及PaeHD组与lfR组相比,心肌梗死体积减小,凋亡细胞减少;心肌组织HMGBl水平明显减少。其中PaeHD组更明显。结论:丹皮酚能够减轻大鼠心肌缺血再灌注损伤,其机制可能与减轻大鼠HMGBl表达有关。

Abstract:Objective:To investigate the paeonol~ influence on HMGB1 expression of myocardial ischemia reperfusionin rats. Methods :40 SD rats were randomly divided into 4 groups:sham operation group (Sham group), ischemia reperf usion group( I/R group) ,low dose of paeonol group( PaeLD group)and high dose of paeonol group( PaeHD group). Rat myocardial infarction volume was detected by TI'C staining method, myocardial ischemia necrosis apoptosis by Tunnel method,while HMGB1 expression in ischemia necrosis tissue by immunoblotting. Results:Compared with LCR group, the volume of myocardial infarction,apoptotic cells number and serum HMGB1 levels in PaeLD and PaeHD group were de creased significantly( P 〈 0.01 ) while the PaeHD group was more obvious. Conclusion : Paeonol can reduce rats myocardial ischemic reperfusion injury, the mechanism may be related to the decrease of HMGB1 mediated inflammatory response in myocardial ischemia reperfusion.
