人参皂甙Rb1和Rg1对体外培养大鼠胚胎生长发育的影响

摘    要:目的探讨人参皂甙Rb1和Rg1对体外培养大鼠胚胎的发育毒性。方法采用植入后全胚胎培养模型,将9.5d龄的SD大鼠胚胎在含有不同浓度人参皂甙Rb1和Rg1的大鼠即刻离心血清中旋转培养48h(Rb1终浓度分别为0、5、15、30、40、50mg/L;Rg1终浓度分别为0、10、30、50mg/L),观察人参皂甙Rb1和Rg1对大鼠胚胎生长发育和组织器官形态分化的影响。结果随着人参皂甙Rb1和Rg1浓度的增加,胚胎生长发育指标得分和胚胎总形态学得分均降低,人参皂甙Rb1浓度≥30mg/L时,与对照组相比,差异有显著性(P<0.05),胚胎体位、心脏和视觉系统得分亦显著减低;当浓度达到50mg/L时,前后肢芽开始受到影响;Rg1浓度≥10mg/L时,胚胎生长发育指标得分与对照组相比,差异有显著性(P<0.05);当浓度≥30mg/L时,前后肢芽得分显著降低(P<0.05);浓度达到50mg/L时,体位、心脏和体节受到影响。结论人参皂甙Rb1和Rg1在较高浓度时对体外培养的大鼠胚胎有一定的毒性作用。

Abstract:Objective To evaluate the developmental to x icity of ginsenoside Rb1 and Rg1 on rat embryos cultured in vitro. M ethods Whole embryo culture system was used in this research. Embryo s were exposed to various concentrations of ginsenoside Rb1 and Rg1, and scored for growth and development at the end of the 48 h culture period. Re sults With the increase in Rb1 concentration, the total morphologica l score decreased. The whole embryonic growth was affected, represented as reduc ed crown rump length, head length and diameter of visceral yolk sac from 30 mg /L . At the same concentration, developments of flexion, heart and optic system were influenced. Limb buds were relatively late and affected at 50 mg/L. With t he concentration of Rg1 rising, the whole embryonic growth was affected from 10 mg/L and the total morphological score remarkably decreased form 30 mg/L. There was also significantly reduced score in limb buds at 30 mg/L. The effects on fle xion, heart and somite were observed at 50 mg/L. Conclusion At higher concentration, ginsenoside Rb1 and Rg1 has developmental toxicity on rat embryos.
