IL-21对Raji细胞增殖抑制和凋亡诱导作用的实验研究

摘    要:目的 探讨IL-21对淋巴瘤Raji细胞增殖抑制和凋亡诱导的作用及其机制。方法 MTT比色法检测观察IL-21对Raji细胞增殖的影响;流式细胞仪观察24h后IL-21对细胞凋亡的影响;RT-PCR检测Livin及caspase-3mRNA水平,观察IL-21对基因表达的影响。结果 IL-21对Raji细胞增殖有抑制作用,呈时间和剂量依赖性(P〈0.05);0、10、100μg/L组细胞凋亡率分别为7.26%、15.12%、22.57%,药物组与对照组比较,差异有统计学意义(P〈0.01或〈0.05);随着IL-21浓度升高,LivinmRNA的表达水平逐步下降,caspase-3mRNA表达逐步升高(P〈0.05)。结论 IL-21可以抑制淋巴瘤细胞的增殖和促进细胞的凋亡,可能与调节Livin、caspase-3基因表达有关。

Abstract:Objective To study the effects of Interleukin-21 (IL-21) on the growth and apoptosis of the Malignant Lymphoma's Cell line Raji and the probable meehanism involved. Methods MTT was used to investigate the growth of Raji cells, flow cytometry (FCM) was used to test the effects of IL-21 on apoptosis of Raji cells; The mRNA expression of Livin and caspase-3 in Raji cells treated with different concentracfion of IL-21 were examined by RT--PCR. Results The growth of Raji cells were inhibited by IL--21 in the time--dose dependent manner (P 〈 0.05) ; FCM showed that the apop- tosis rates of cells in 10ug/L (15.12%) and 100 ug/L IL-21 group (22.57%) were higher than those in 0ug/L IL-21 group (7.26 %), with significant difference (P 〈 0.01 or 〈 0.05) ; the expression rates of Livin mRNA were decreased by the treatment with the increasing concentrations of IL-- 21, while caspase--3 increased (P 〈 0.05). Conclusions IL--21 can restrain the human lymphoma's cell growth and induce cell apoptosis, which may be related to regulate the expressions of Livin and caspase--3.
