加减地黄饮子预处理对局灶性脑缺血大鼠脑组织神经生长因子表达的影响

目的观察加减地黄饮子预处理对局灶性脑缺血大鼠脑组织神经生长因子(NGF)表达的影响及其可能的神经元保护机制。方法采用栓线法制作大鼠局灶性脑缺血损伤模型(MCAO)。将120只Wistar大鼠随机分成6组:空白组、假手术组、模型组、中药高剂量组、中药低剂量组、西药对照组,每组20只。连续灌胃7 d后,进行手术操作。HE染色法观察大鼠脑组织病理学改变;免疫组织化学法观察大鼠脑组织NGF的表达。结果低剂量、高剂量加减地黄饮子均能保护梗死区神经细胞,细胞结构较完整;增强梗死区脑组织NGF的表达。结论加减地黄饮子预处理可对局灶性脑缺血大鼠脑组织神经元起保护作用,其脑保护作用机制可能与调节大鼠脑组织NGF表达及保护梗死区神经细胞结构有关。

Influence of Dihuangyinzi decoction pretreatment on expression of nerve growth factor in brain tissue of rats with focal cerebral ischemia

Objective It is to observe the influence of Dihuangyinzi decoction pretreatment on the expression of nerve growth factor(NGF) in brain tissue of rats with focal cerebral ischemia and the possible neuron protection mechanism.Methods Focal cerebral ischemia rat models were made with line fasten method.120 Wistar rats were randomly divided into blank group,sham operation group,model group,traditional Chinese medicine high dose group, traditional Chinese medicine low dose group and western medicine control group. There were 20 rats in every group.After continuously intragastric administration for seven days,the operations were done.The pathology changes in brain tissue of rats were observed with HE staining method.The expressions of NGF in brain tissue of rats were observed with immunohistochemical method.Results Low dose and high dose Dihuangyinzi decoction both could protect the nerve cell in infarct region.The cellular structures were comparatively complete.The expressions of NGF in brain tissue of infarct region were strengthened.Conclusion Dihuangyinzi decoction pretreatment can protect the neuron in brain tissue of rats with focal cerebral ischemia.The brain protection mechanism may be concerned with regulating the expressions of NGF in brain tissue of rats and protecting the structure of nerve cell in infarct region.
