清金补肺汤对博莱霉素致大鼠肺纤维化干预作用的研究

目的探讨清金补肺汤对实验性大鼠肺纤维化的保护作用及其相关机制。方法将45只清洁级Wistar雄性大鼠随机分为盐水对照组15只、BLM模型组15只和治疗组15只。模型组和治疗组采用一次性气管内注入BLM诱导大鼠肺纤维化模型,盐水对照组在相同条件下给予等量的生理盐水气管内滴注。治疗组造模后第2天起给予清金补肺汤2 mL灌胃,模型组和盐水对照组给予等量的蒸馏水灌胃。各组分别于造模后第7天、第14天和第28天随机处死5只大鼠,留取血清及肺组织。用酶联免疫吸附测定法检测血清中TNF-α的含量;应用HE染色和Masson染色观察肺泡炎和肺纤维化的动态变化。结果病理切片可见模型组造模成功。模型组大鼠血清中的TNF-α含量较盐水对照组各时间点明显升高(P均<0.01);与模型组比较,治疗组各时间点血清中TNF-α含量均明显降低(P均<0.01)。结论清金补肺汤可降低博莱霉素所致肺纤维化大鼠血清中TNF-α的含量;早期应用清金补肺汤干预治疗博莱霉素致肺纤维化大鼠,可减缓其肺纤维化进程。

The intervention effect of Qingjin Bufei decoction on rats with lung fibrosis induced by bleomycin

Objective It is to explore the protection and the relevant mechanism of Qingjin Bufei decoction on lung fibrosis process in rats.Methods 45 clean level Wistar male rats were randomly divided into three groups: salt water group,bleomycin(BLM) model group and treatment group.There were 15 rats in each group.Rats in BLM model group and treatment group were given BLM one-time injected into the windpipe which could establish lung fibrosis model.Rats in salt water group were given saline water at the same conditions.Rats in treatment group were given Qingjin Bufei decoction 2 mL/kg by intragastric administration on the next day after the models were established.Rats in BLM model group and salt water group were given the equal amount of distilled water intragastric administration.In each group,in 7,14 and 28 days after model established,5 rats were killed randomly and their serum and lung tissue were gotten to determine serum TNF-α,alveolitis and dynamic changes of pulmonary fibrosis were observed by HE stain and Masson HE.Results The model groups were established successful according to pathologic section.TNF-α levels in serum of BLM model group compared with that in salt water group were significantly higher at each time point,(P<0.01).Compared with BLM model group,TNF-α levels in serum in treatment group were significantly lower at each time point(P<0.01).Conclusion Qingjin Bufei decoction can reduce TNF-α serum levels in rats with fibrosis induced by BLM.Early intervention of Qingjin Bufei decoction can slow down the process of pulmonary fibrosis in rats with fibrosis induced by BLM.
