蕈样肉芽肿皮损中DNA含量及细胞增殖活性的检测

摘    要:目的:研究蕈样肉芽肿(MF)皮损中DNA含量、细胞周期分布与分期的关系,以探讨DNA倍体及细胞增殖活性在该病早期诊断、治疗及预后判断方面的作用。方法:应用流式细胞仪检测38例蕈样肉芽肿皮肤损害石蜡包埋组织中的DNA含量及细胞周期分布。结果:38例蕈样肉芽肿皮损石蜡包埋组织中异倍体率、S期细胞比率(SPF)及细胞增殖指数(PI)随病情的进展而增高。红斑期与斑块期、斑块期与肿瘤期的SPF值及红斑期与斑块期PI值之间有统计学差异(P<0.001);斑块期与肿瘤期PI值之间无统计学差异(P>0.05)。结论;蕈样肉芽肿皮损组织中的DNA倍体及细胞增殖活性有可能成为该病临床诊断及预后判断的重要因素。

Abstract:Objective:To investigate the relationship between DNA content,cell cycle distri-bution in paraffin embedded tissue of mycosis fungoides(MF)and stage of the disease and the role ofthem in early diagnosis,treatment and prognosis.Methods:DNA content and cell cycle distribution inparaffin embedded tissue of 38 patients with MF are measured by flow cytometry.Results:The rateof aneuploidy,S-phase fraction(SPF)and proliferative indices(PI)were increased with progression ofMF.There were significant differences in SPF between the stages of erythema and plaque,plaque andneoplasm(P<0.001).There was a significant difference in PI between the stages of erythema andplaque(P<0.001),but there was no significant difference in PI between the stage of plaque and neo-plasm(P>0.05).Conclusion:DNA ploidy and cell proliferative activity may be important factors in diag-nosis and prognosis of MF.
