全反式维甲酸可增强大鼠系膜细胞uPA和MMP—2表达

摘    要:目的 研究全反式维甲酸对体外培养大鼠系膜细胞uPA和MMP-2表达的影响,探讨全反式维甲酸抗肾小球硬化的机制。方法 分别应用Northern blot、Western blot和酶谱法检测全反式维甲酸对培养大鼠系膜细胞uPA和MMP-2表达的影响。结果 ATRA可促进系膜细胞uPA和MMP-2蛋白表达,提高其酶活性,并增强MMP-2mRNA表达。结论 ATRA可增强大鼠系膜细胞的uPA和MMP-2表达,为进一步阐明ATRA拮抗肾小球硬化机制提供了直接的实验依据。

Abstract:Purpose To study the effects of all trans retinoic acid on the expression of uPA/MMP?2 in cultured rat mesangial cells, and to elucidate the mechanism by which all trans retinoic acid prevents age related progressive glomemular sclerosis. Methods The mRNA, protein and enzymatic activity of uPA/MMP?2 were detected by Northern blot analysis, Western blot analysis and Zymography assay, respectively. Results All trans retinoic acid upregulated the expression of uPA/MMP?2 protein, enhanced the enzymatic activity of uPA/MMP?2. Thus all trans retinoic acid elevated the level of MMP?2 mRNA in cultured mesangial cells. Conclusions All trans retinoic acid upregulated the expression of uPA/MMP?2 in cultured rat mesangial cells. It is valuable for explaining the role of all trans retinoic acid in preventing age related glomerulsclerosis.
