脂质体包裹可溶性蛋白质抗原诱导特异性CTL的研究

摘    要:目的:搪塞用可溶性蛋白质抗原诱导特异性CTL的途径。方法:用冻融-超声法制备包裹可溶性鸡卵蛋白(OVA)和CA-Hb3蛋白质抗原的阳离子脂质体,并借助脂南体将抗的导入C57BL/6小鼠脾细胞胞浆内,以此细胞经尾静脉免疫C57BL/6小鼠,取体内初致敏脾细胞,在体外以渗透方式导入OVA或CA-Hb3抗原的脾细胞再刺激,诱导特异性CTL,并进行I类途径阻断实验和MHCI类分子中和实验,结果:脂质体成功

Abstract:Objective:To determine whether soluble protein antigen could induce specific CTL response. Methods:Soluble protein antigen OVA and CA Hb3 were encapsulated in cationic liposomes by thaw freeze ultrasonic method and introduced into mice splenocytoplasms, Then injected into C57BL/6 mice through their tail veins,then primed the splenocytes in vivo restimulated in vitro by splenocytes loaded antigen with osmotic means for induction of specific CTL. Blockage of class I pathway and neutrolization of MHC I molecule were also applied. Results:The antigens encapsulated in cationic liposomes and introduced into the cytoplasms were observed under electromicroscope, immunohistochemical assay proved the presence of expression of the antigen on the cells, and cytotoxic ity assay showed that CTL were antigen specific and MHC restricted, Blockage of class I pathway proved that the antigens were presented via class I pathway, neutralization of MHC I molecule indicated the recognition and killing of the CTL were related to MHC I molecule. Conclusions:Soluble protein antigen can be encapsulated in cationic liposomes and introduced into the cytoplasms, which an present through class I pathway, and induce specific CTL responses.
