萆薢牛膝总皂苷对尿酸钠诱导的大鼠急性痛风性关节炎的防治作用

目的:研究萆薢牛膝总皂苷对大鼠急性痛风性关节炎的防治作用及机制。方法:大鼠ig总皂苷高、中、低剂量(200、60、20mg/kg)、吲哚美辛组(5mg/kg),每天1次,连续5天。采用微晶型尿酸钠关节腔内注射诱导大鼠急性痛风性关节炎,测定关节肿胀度、关节浸出液中炎症因子、细胞因子水平,并进行病理学检查。结果:模型组大鼠关节明显肿胀,活动减少,右后肢弯曲或抬起,肿胀关节浸出液中NO、TNF-α、PGE2、IL-1β、IL-6和IL-8水平显著增高,关节滑膜多核白细胞、单核细胞浸润,并可见针状尿酸盐结晶沉着。总皂苷200、60mg/kg组大鼠关节肿胀度明显降低,关节及周围组织病理学病变显著减轻。200mg/kg组大鼠步态明显好转,关节浸出液中NO、TNF-α、PGE2和IL-6、IL-1β均显著降低,60mg/kg组大鼠肿胀关节浸出液中TNF-α、PGE2显著降低。结论:总皂苷对大鼠急性痛风性关节炎有明显的防治作用,其机制可能是抑制炎症细胞趋化和激活,抑制炎症因子与细胞因子的合成与释放。
