糖尿病模型大鼠表征及其证候动态演变研究

摘    要:目的:观察2型糖尿病大鼠的表征变化及证候动态演变。方法:将38只SD大鼠随机分为正常对照组和模型组。模型组给予高脂饲料喂养3周后皮下注射甲状腺素(T4)0.2mg/(kg.d)1周,后腹腔注射链脲佐菌素(STZ)35mg/kg。连续采集两组大鼠的表征及体重、24h饮水量、进食量;并测定空腹血糖(FBG)、空腹胰岛素(FIN)、总胆固醇(TG)、甘油三酯(TC)、葡萄糖耐量(OGTT)。结果:模型组大鼠FBG、TG、TC、OGTT、24h饮水量、进食量均明显升高;体重均明显降低。模型组大鼠在注射STZ后3~7周出现:活动次数增加、背温升高、各自爬卧、舌色鲜红、舌干少津、大便干硬比率升高;在注射STZ后9~11周出现:活动次数减少、舌色鲜红、舌干少津、爪色青紫比率升高。结论:判定模型大鼠在STZ注射后3~7周为阴虚内热证型,在STZ注射后9~11周为气阴两虚证型。

Abstract:Objective:To explore the characteristic and syndrome dynamic succession of type- 2 diabetic rats. Methods :38 rats were randomly divided into two groups in terms of control group and model group. The rat models were injected T4 0.2mg/( kg · d) for a week. The rat models were built up with STZ(35mg/kg) once intra -peritoneally after feeding with high fat chow for 4 weeks. The characteristic was collected successively, as well as the amount of food, water, and weight. The OGTr, FBG, FIN, TG, TC were also detected. Results : The amount of food , water, and OGTT, FBG, TG, TC were obviously increased. The amount of weight was obviously decreased. After 3 - 7 weeks of injection of STZ, the rate of characteristics of increase in times of activity, in- crease in temperature of back,lying down respectively, bright redness of tongue, dry tongue, and dry stool were increased. After 9 - 11 weeks of injection of STZ, the rate of characteristics of decrease in times of activity, bright redness of tongue, dry tongue, and cyanosis of claws were increased. Conclusion : The rat models were syndrome of yin deficiency with internal heat in time of 3 - 7 weeks after injection of STZ , and were syndrome of dual deficiency of qi and yin in time of 9 - 11 weeks after injection of STZ.
