HPLC法测定麸炒山药饮片中尿囊素的含量

摘    要:目的建立麸炒山药饮片中尿囊素含量测定的方法,测定不同产地麸炒山药饮片中尿囊素的含量。方法采用ODS-C18柱(250 mm×4.6 mm,5μm);流动相:甲醇-磷酸二氢钾水溶液(2∶98);流速0.8 mL/min;柱温:30℃;检测波长:224 nm。结果尿囊素在0.144~2.312μg范围内线性关系良好,r=0.9999;尿囊素的平均回收率为99.05%,RSD=2.99%(n=9)。结论该方法可为麸炒山药饮片的质量控制提供依据,不同产地麸炒山药中尿囊素含量差异较大。

Detection of Allantoin in Bran-baked Sliced Rhizoma Dioscoreae with HPLC

Abstract:OBJECTIVE To establish a method for the determination of allantoin and determine content of allantoin in Rhizoma Dioscoreae Preparata from different producing areas.METHODS Separation and determination of allantoin were accessed by using an ODS-C18 column and a mobile phase8 of methanol - potassium dihydrogen phosphate aqueous solution at a ratio of 2:98 (v/v). The flow rate was 0.8 mL/min, temperature at 30℃ and detection wavelength at 224 nm. RESULTS A good linearity of allantoin was achieved in the range of 0.144 - 2.132 μg, r= 0.9999; the mean value of recovery is 99.05%, RSD is 2.99%. CONCLUSIONS The method in this paper could provide an evidence for quality control of Rhizoma Dioscoreae Preparata and there is comparatively large difference of content of allantoin in Rhizoma Dioscoreae Preparata from different producing areas.
