Abstract:Objective:To establish a testing method for determination of diosgenin content in Dioscorea nipponica Makino by PHLC-ELSD.Methods:Agilent Eclipse C18(150mm×4.6mm,5μm)column and evaporated light scattering detector(N2 as carrier gas,pressure:3.5bar,drift tube temperature:70℃) were used in this study.Mobile phase was methanol-water(95:5) at 40℃,flow rate was 1.0ml/min.Results:Diosgenin had better linear rate at 2.04-0.255mg/ml(r=0.9996),the average recovery was 98.9%(RSD=1.4%);and this PHLC-ELSD method had better repeatability.Conclusions:This method for detecting diosgenin content in Dioscorea nipponica Makino by PHLC-ELSD is specific,sensitive and accurate,can be acted as effective method for quality control of Dioscorea nipponica Makino.
