原发性胆汁性肝硬化患者外周血单个核细胞中Tim-3 mRNA表达增高及其意义

摘    要:为研究Tim-3是否参与了原发性胆汁性肝硬化(PBC)的发病机制,我们采用实时荧光定量逆转录-聚合酶链反应(RT-PCR)技术检测了38例PBC患者及30例健康者外周血单个核细胞(PBMC)中Tim-3mRNA的相对表达量,并分析Tim-3mRNA表达与PBC患者Mayo危险评分和碱性磷酸酶(ALP)之间的关系。结果表明,PBC患者外周血PBMC中Tim-3mRNA表达较健康对照组明显增高(P<0.01),且与Mayo危险评分呈正相关(r2=0.31,P<0.01),与血清ALP水平呈负相关关系(r2=0.37,P<0.01)。本研究表明Tim-3可能参与了PBC的发病机制,同时还是PBC的潜在标志物。

Abstract:To investigate whether Tim-3 was involved in the pathogenesis of primary biliary cirrhosis(PBC),the relative expression of Tim-3 mRNA in the peripheral blood mononuclear cells(PBMC) of 38 PBC patients and 30 healthy volunteers were delermined by RT-PCR.For PBC patients,the relationship between Tim-3 mRNA and serum alkaline phosphatase(ALP) activity and Mayo risk score were analyzed.Increased Tim-3 mRNA expression,which positively correlated with Mayo risk score(r=0.31,P<0.01) and negatively correlated with serum ALP activity(r=0.37,P<0.01),was observed in PBC patients.These results indicated that Tim-3 may be involved in the pathogenesis of PBC and could be regarded as a biomarker for PBC.
