20(S)-原人参二醇对人前列腺癌细胞凋亡的诱导作用

摘    要:目的:研究20(S)-原人参二醇对人前列腺癌细胞凋亡的影响。方法:采用台盼蓝染色、噻唑蓝(MTT)法、Hochest33258染色和DNA Ladder方法分别测定20(S)-原人参二醇对DU145和PC3细胞凋亡诱导作用。结果:20(S)-原人参二醇可明显降低DU145和PC3细胞存活率和细胞活性,IC50分别为38.0μmol/L和28.6μmol/L。20(S)-原人参二醇处理后DU145和PC3细胞核出现典型的凋亡形态学改变,琼脂糖电泳呈现明显的DNA Ladder条带。结论:20(S)-原人参二醇可有效诱导人前列腺癌DU145和PC3细胞的凋亡并有剂量依赖关系。

Apoptosis effects of human prostate cancer cells induced by 20(S)-protopanaxadiol
Abstract:Objective:To study the effects of 20(S) -protopanaxadiol(PPD) on the apoptosis of human prostate cancer cells.Methods:DU-145 and PC-3 human prostate cancer cell lines were treated with PPD.Cell growth rate was determined with Typan Blue and MTT assay.The effects of PPD on apoptosis rate were observed by the Hochest 33258 dye and DNA ladder.Results:20(S)-Protopanaxadiol could inhibit the proliferation of DU-145 and PC-3 cells,and IC50 values were 38.0μmol/L and 28.6μmol/L.The morphological changes were observed in the nucleus of DU-145 and PC-3 cells treated by PPD.The DNA Ladders bands were detected by 1% agrose gel electrophoresis.Conclusion:20(S)-Protopanaxadiol could induce DU-145 and PC-3 cell apoptosis in a dose dependent manner.
