丙型肝炎病毒NS3区C末端HLA-11限制性CTL表位的作用

目的 研究丙型肝炎病毒（ｈｅｐａｔｉｔｉｓ Ｃ ｖｉｒｕｓ，ＨＣＶ）非结构３区（ＮＳ３）Ｃ末端ＨＬＡ－１１限制的细胞Ｔ细胞（ＣＴＬ）表位在ＨＣＶ感染中的作用。方法 血清学方法检测３例患的ＨＬＡ分型，他们均存在ＨＬＡ－Ａ１１表型，以ＨＬＡ－１１结合基序为依据，预测了ＨＬＡ－１１限制的ＣＴＬ表位。２例慢性患５ａ内３个时间点和３ａ内２个时间点及转阴患的血清样本，用反转录ＰＣＲ扩增出ＨＣＶ基因片段，

HLA-A11 restricted CTL epitopes in C-terminal of HCV NS3 region has no effects on HCV infection

AIM To investigate the effect of HLA-A11 restricted CTL epitope in the C-terminal of HCV NS3 region on HCV infection. METHODS HLA typing was determined with serological method in three HCV patients.Based on this, the HLA restricted CTL epitopes were predicted by HLA binding motif. The genes of serum samples were amplified by PCR, then sequenced and translated into proteins. RESULTS No obvious changes were observed in predicted CTL epitopes in two chronic HCV patients during 5 and 3 years,but in the patient recoverying from HCV infection,there was a changed HLA-A11 restricted CTL epitope(sequence IILTHPVTK) which T lymphocyte proliferation experiment proved was not an HLA-A11 restricted CTL epitope. CONCLUSION HLA-A11 restricted CTL epitope in the C-terminal of HCV NS3 protein may have no relationship with the recovery of HCV infection.
