高脂高糖饮食对自发性高血压大鼠内皮素表达及肾功能的影响

摘    要:目的探讨高脂高糖饮食对自发性高血压大鼠(SHR)内皮素1表达和肾功能的影响。方法 24只6周龄雄性SHR随机分为高脂高糖组和普通饲料组,每组12只。观察高脂高糖饮食对体质量、血糖、血脂、胰岛素、内皮素1和肾功能等的影响,计算胰岛素抵抗指数(HOMA-IR)、肌酐清除率(Ccr)、尿白蛋白/肌酐比值(ACR)的变化。提取肾脏总RNA,实时定量RT-PCR检测内皮素1mRNA,HE染色观察肾脏的病理变化。结果与普通饲料组比较,高脂高糖组大鼠干预后第12周体质量增加24%,TC、TG水平、胰岛素及HOMA-IR升高,内皮素1增高[(45.0±0.2)ng/L vs(25.0±0.1)ng/L];Ccr降低[(0.3±0.2)ml/min vs(0.6±0.1)ml/min],差异有统计学意义(P<0.05,P<0.01)。ACR升高(P<0.05);肾脏内皮素1mRNA增高(197.9±22.2)vs(100.3±11.4),差异有统计学意义(P<0.05)。结论长期高脂高糖饮食可加重SHR肾损害,其机制可能与胰岛素升高、HOMA-IR增加及肾脏内皮素1mRNA表达上调相关。

Effect of high fat and glucose diet on endothelin 1 expression and renal function in spontaneous hypertensive rats
Abstract:Objective To study the effect of high fat and glucose diet on endothelin 1(ET1)expression and renal function in spontaneous hypertensive rats(SHR).Methods Twenty-four male SHR aged 6weeks were randomly divided into high fat and glucose diet group and normal chow diet group(12in each group).The effect of high fat and glucose diet on body weight,blood glucose and blood lipid level,serum insulin and ET1levels and renal function was observed.The insulin resistance index(HOMA-IR),creatinine clearance rate(Ccr)and urine albumin/creatinine ratio were calculated.RNA was isolated from the kidney.ET1mRNA expression was detected by RT-PCR.Glomerulosclerosis index(GSI)and tubulointerstitial fibrosis(TIS)were observed with HE staining.Results The body weight was 24% higher,the average serum TC and TG levels were higher,the serum insulin level and HOMA-IR were 1.83-fold and 1.85-fold higher,the serum ET1level was higher(45.0±0.2ng/L vs 25.0±0.1ng/L)while the serum Ccr level was lower(0.3±0.2 vs 0.6±0.1ml/min),the ACR was higher,and the ET1mRNA expression level was higher(197.9±22.2 vs 100.3±11.4)in high fat and glucose diet group than in normal chow diet group(P<0.05,P<0.01).Conclusion Long-term high fat and glucose diet aggravates renal dysfunction in SHR by increasing the insulin level and HOMA-IR and upregulating the ET1 mRNA expression.
