子宫内膜癌中PTEN蛋白、HER-2蛋白的表达及意义

目的检测子宫内膜癌组织中PTEN蛋白及HER-2蛋白表达并探讨其意义。方法采用免疫组织化学法检测60例子宫内膜癌、32例正常子宫内膜组织中PTEN蛋白及HER-2的表达。结果子宫内膜癌组织中PTEN蛋白为缺失表达,高于正常子宫内膜组织(P<0.05),而且PTEN表达在G1级肿瘤高于G2、G3级(P均<0.05),PTEN蛋白缺失率与肿瘤组织类型有关(P<0.05),与肌层浸润、淋巴结转移及病理分期无明显关系(P均>0.05)。子宫内膜癌组织中HER-2蛋白表达为高表达,HER-2的表达与肿瘤组织学分级、病理学分期以及肿瘤浸润子宫肌层深度均显著相关(P均<0.05),其表达与组织学类型、有无淋巴结转移均无关(P均>0.05)。HER-2与PTEN的表达呈明显负相关。结论 PTEN表达缺失与临床病理参数无关,蛋白表达缺失常发生于细胞分化较差的子宫内膜癌。PTEN蛋白表达缺失与HER-2表达水平有关。

Eexpression of PTEN protein and HER-2 protein and its significance in endometrial carcinoma

Objective It is to detect the expression of PTEN protein and HER-2 protein in endometrial carcinoma and to explore its significance.Methods The expression of PTEN protein and HER-2 in 60 cases of endometrial carcinoma tissue and 30 cases of normal endometrial tissue were detected by immunohistochemical assay.Results PTEN protein expression in endometrial carcinoma tissue was loss,and was higher than that in normal endometrial tissue(P<0.05),and the PTEN expression in G1 level was higher than that of G2,G3 tumor(P<0.05).PTEN protein expression loss rate was related with tumor tissue types(P<0.05),but was not related with myometrial invasion,lymph node metastasis and pathological(P>0.05).HER-2 protein had a high expression in endometrial carcinoma,and its expression was significantly related with tumor histological grade,pathological stage and tumor infiltrating depth in myometrium(P<0.05),but was not associated with histologic types and lymph node metastasis that existed or not(P>0.05).There was a significantly negative correlation between HER-2 and PTEN expression.Conclusion PTEN expression is not related with clinical pathology parameters,and the protein loss often occurs in poorly differentiated endometrial cancer cells.Expression loss of PTEN protein is related with the expression levels of HER-2.
