摘    要:目的:探讨周围神经损伤后神经型一氧化氮合酶羧基末端PDZ配体(carboxy temlinal PDZ ligand of neuronal nitric oxide synthase,CAPON)的表达变化与定位。方法:利用实时荧光定量PCR(real-time fluorescence quantitative PCR,FQPCR)、原位杂交与间接免疫荧光相结合的方法,研究大鼠坐骨神经损伤后CAPON mRNA表达变化及其定位。结果:CAPON mRNA主要分布在正常和损伤坐骨神经的雪旺细胞(Schwann cells,SCs)中,损伤后表达增多,分别在神经夹伤后2周以及切断后1周的近、远侧断端中出现表达高峰。原代培养的SCs中也检测到CAPON的表达,其mRNA分布在核周胞质区域。结论:周围神经损伤后引起CAPON mRNA表达上调,增殖的SCs中表达CA- PON可能对神经损伤后冉生修复发挥一定作用。

Abstract:Objective:To study the localizations and changes of carboxy terminal PDZ ligand of neural nitric oxide syn- thase(CAPON)both in injured peripheral nerves.Methods:Real-time fluorescence quantitative PCR(FQ-PCR),as well as a combination of in situ hybridization(ISH)and indirect immunofluorescence were used to detect the localiza- tions and changes of CAPON mRNA.Results:It was found that CAPON mRNA was mainly localized in Schwann cells (SCs)of the normal sciatic nerve,however,increased during injury,and reached its peak at 2 w in crushed sections of the sciatic nerves and at 1 win both approximate and distal stumps of transectcd nerves.The expression of mRNA for CAPON was confirmed in primary SCs culture,which principally lied in perinuclear plasm.Conclusion:The upregula- tion of mRNA for CAPON in SCs may be involved in axonal regeneration after periphery nerve injury.
