双侧颈总动脉缺血再灌注后大脑皮质一氧化氮合酶和生长抑素的表达

摘    要:目的:探讨脑缺血再灌注后大脑皮质一氧化氮合酶(NOS)和生长抑素(SS)的基因表达及其意义。方法:取Wistar大鼠,缺血再灌注分6、24和72 h组。测试大脑皮质一氧化氮合酶和生长抑素蛋白表达水平。结果:NOS缺血再灌注6、24 h组和对照组相比阳性神经元数明显升高;缺血再灌注72 h与6、24 h组相比阳性神经元数明显下降;SS缺血再灌注6、24、72 h组与对照组相比阳性神经元数明显下降;缺血再灌注72 h组比缺血再灌注6 h组下降更低。结论:NOS和SS通过多种途径参与了脑缺血再灌注后细胞损伤的发生。

Abstract:Objective:To investigate the gene expression of nitric oxide synthase (NOS) and somatostatin (SS) in the cerebral cortex after occlusion of bilateral common carotid artery and reperfusion in rats.Methods:Forty healthy Wist- ar rats were randomly divided into 4 groups:6 h,24 h and 72 h after cerebral ischemia-reperfusion groups,and the con- trol group.NOS and SS expressions in the cerebral cortex were observed by NADPH-d histochemistry and immunohis- toehemistry.Results:(1)The numbers of NOS positive neurons in 6 h and 24 h groups were significantly higher than that in the control group,whereas there was no significant difference between 72 h and the control group and between 6 h and 24 h groups,and the number in 72 h group was significantly smaller than those in 6 h and 24 h groups.(2) The numbers of SS-irnmunoreactive neurons in 6 h,24 h and 72 h groups were significantly smaller than that in the control group,whereas the number in 72 h group was even smaller than that in 6 h group,and no difference was found between 6 h and 24 h groups and between 24 h and 72 h groups.Conclusion:NOS and SS may be invovled in neuronal injury after cerebral ischemia-reperfusion through multiple ways.
