摘    要:目的:研究p27~(kip1)和Skp2在正常和坐骨神经夹伤后大鼠脊髓中的表达变化。方法:Westem印迹法,免疫组织化学,免疫荧光双标。结果:Western印迹结果显示坐骨神经夹伤后,脊髓中p27~(kip1)表达下降后又有所恢复,并且p27~(kip1)和Skp2在脊髓中的表达变化呈负相关。免疫组织化学及免疫荧光双标显示,p27~(kip1)和Skp2在正常和损伤后的脊髓神经元和胶质细胞中都有表达。坐骨神经夹伤后p27~(kip1)和Skp2在脊髓腹角阳性细胞数目变化呈负相关。结论:坐骨神经损伤后p27~(kip1)。和Skp2在脊髓腹角神经元及其周围的胶质细胞表达变化相关,对研究脊髓损伤和修复的分子机制有重要意义。

Abstract:Objective:To investigate expressions of p27~(kip1) and Skp2 in normal and injured rat spinal cords.Methods: Western blot,immunohistochemistry and double immunolabelling were used.Results:Expressions of p27~(kip1) and Skp2 were detected in rat spinal cords.The level of p27~(kip1) in the spinal cord was declined after sciatic nerve crush,and then it was rescued.And the changes of p27~(kip1) and Skp2 in rat spinal cords after sciatic nerve injury were inversely correla- ted.It was showed that p27~(kip1) and Skp2 were expressed in the neurons and gliocytes.Statistical analysis indicated that there was negative correlation between the numbers of p27~(kip1) and Skp2 positive cells in the ventral horn after sciatic nerve injury.Conclusion:After sciatic nerve injury,the expressions of p27~(kip1) and Skp2 were correlated tightly in the neurons and the glial cells around in the ventral horn of the spinal cord,which is of great significance for the investiga- tion of the molecular mechanism of renovation of the spinal cord after peripheral nerve injury.
