摘    要:目的:研究坐骨神经损伤后Roundabout 2(Robo2)在成年大鼠背根节和脊髓的表达变化。方法:健康成年雌性SD大鼠坐骨神经切断后分别存活3~28d,取其L_(4~6)背根节(DRG)和脊髓;利用RT-PCR和免疫组织化学技术检测Robo2在上述组织中的表达变化。图像分析技术对阳性细胞的灰度值进行测定。结果:正常DRG感觉神经元表达Robo2 mRNA和蛋白质,脊髓前角运动神经元不表达。坐骨神经切断后3 d DRG内Robo2表达增加,7~14 d达高峰,21~28 d恢复到正常水平。结论:坐骨神经切断可导致DRG内Robo2的表达上调,可能与早期的感觉轴突再生有关。

Abstract:Objective:To investigate the expression changes of Roundabout2(Robo2) in the dorsal root ganglion (DRG) and spinal cord of rats after sciatic nerve transection.Methods:Adult female Sprague-Dawley rats were randomly di- vided into the normal control group and the experimental group.In the experimental group,the right sciatic nerve was transeeted and survived for 3,7,14,21 and 28 days after operation.The changes in the experssion of Robo2 mRNA and protein in L4-6 DRG and spinal cord were detected by RT-PCR and immunohistochemical methods.Quantitative a- nalysis was performed using image analysis system.Results:The sensory neurons in DRG constitutively expressed Ro- bo2 mRNA and protein,but no expression was in spinal cord.After sciatic nerve transection,the expression of Robo2 was up-regulated at day 3,peaked at day 7 and 14,then returned to normal.Conclusion:The expression of Robo2 in DRG can be influenced by peripheral nerve injury,suggesting that Robo2 may be involved in the regulation of sensory nerve regeneration.
