摘    要:[摘要] 目的:探究雷公藤甲素对人子宫颈微血管生成的抑制作用,并探讨其作用机制。方法:选择人子宫颈微血管内皮细胞(HCerMEC)作为研究对象,体外培养过程中分别给予0、5、10和20、40 ng/ml的雷公藤甲素处理,采用CCK-8 法测定其对细胞增殖的影响,transwell实验检测细胞的迁移能力,western blotting 检测细胞血管内皮生长因子(VEGF)表达的变化。结果:雷公藤甲素抑制HCerMEC的增殖活性和细胞迁移能力,并呈剂量依赖关系(P<0.05)。雷公藤甲素能够抑制HCerMEC细胞内VEGF的表达,药物浓度越高抑制作用越强。结论:雷公藤甲素可以抑制HCerMEC细胞增殖和迁移活性,该作用与其抑制VEGF表达有关。

Abstract:[Abstract] Objective: To investigate the inhibitory effect of Triptolide on vasculogenesis of human cervical microvascular endothelial cells, and to explore the mechanism. Methods: Human cervical microvascular endothelial cells (HCerMECs) were used as research subject,and treated with different concentrations of Triptolide (0, 5, 10, 20 and 40 ng/ml) in vitro. The effect of Triptolide on cell proliferation was determined by CCK-8, the cell migration ability was detected by Transwell assay while the expression of vascular endothelial growth factor (VEGF) was examined by western blotting. Results: Triptolide inhibited the proliferation and migration of HCerMECs in a dose-dependent manner (P<0.05). In addition, Triptolide could inhibit the expression of VEGF in HCerMECs in a concentration-dependent manner. Conclusions: Triptolide could inhibit the proliferation and migration activity of HCerMECs which is related with the suppression of VEGF expression.
