胰腺癌线粒体DNA突变研究

摘    要:目的研究线粒体DNA(mtDNA)突变在胰腺癌发病中的作用。方法用PCR与直接测序相结合的方法,对比分析2株胰腺癌细胞株(SW1990、JF-305)和1株原代培养的正常胰腺细胞mtDNA D环区的突变位点。结果 2株胰腺癌细胞和1株正常的胰腺细胞的mtDNA D-loop区均存在不同程度的点突变,SW1990共检测到8个突变位点,JF-305共检测9个突变位点。其中73位A-G、16223位C-T和16358位C-T这3个突变位点在2株癌细胞和正常胰腺细胞中均检测到,考虑为多态性变化;16211位C-T和16311位T-C2个相同的突变位点在2株胰腺癌细胞中均检测到,考虑为特征性突变。结论胰腺癌细胞mtDNAD环区具有多态性和突变性,其突变可能与胰腺癌的发生、发展密切相关。

Abstract:Objective To investigate mutation situation of pancreatic cancer cell mitochondria DNA D-loop region.Methods The mutational site of mitochondria DNA D-loop region in two strains of pancreatic cancer cells(SW1990 and JF-305) and one normal pancreas cell cultured primarily were analyzed by PCR and direct sequencing.Results The point mutations in pancreatic cancer cells and normal pancreas cells were all found;among them,8 point mutations in SW1 990 and 9 point mutations in JF-305 were detected.3 point mutations existed in two strains of pancreatic cancer cells and normal pancreas cells with 73 site A-G,16 223 site C-Tand 16 358 site C-T,which were considered as polymorphism;otherwiset,wo same point mutations were detected in two cell strains of pancreatic can-cers with 16 211 site C-T and 16 311 site T-C,which was considered as special mutations.Conclusions Polymorph-ism and special mutations exist in the mitochondrial DNA D-loop region of pancreatic cancer cells.The special mut-ations may be concerned with the occurring and development of pancreatic cancer.
