PDGFBB与VEGF在人脑胶质瘤中的表达及临床意义

摘    要:目的研究血小板源生长因子B链的纯合二聚体(PDGFBB)与血管内皮细胞生长因子(VEGF)在正常脑组织和胶质瘤中的表达及这两种因子与胶质瘤病理参数包括肿瘤的病理分级、大小、复发和预后的关系以及PDGFBB和VEGF之间的相关性。方法应用SABC免疫组织化学染色方法对33例人脑胶质瘤和5例正常脑组织中PDGFBB和VEGF的表达进行检测和统计学分析。结果肿瘤组织中PDGFBB和VEGF高于正常脑组织中PDGFBB和VEGF表达,二者比较,有显著性差异(P<0·01)。肿瘤组织中PDGFBB和VEGF表达呈正相关。肿瘤组织中PDGFBB的表达与肿瘤恶性程度呈正相关关系,与肿瘤大小、复发和预后无相关关系;肿瘤组织中VEGF的表达与肿瘤恶性程度呈正相关关系,与肿瘤大小、复发无相关关系,在预后较差一组的VEGF表达高于预后较好的一组,有统计学意义。结论PDGFBB和VEGF这两种因子在作用机制上可能有密切的联系或存在因果关系,检测PDGFBB和VEGF对判定肿瘤恶性程度有一定的临床意义,检测VEGF对判定肿瘤预后可能有一定的临床意义。

Abstract:Objective To evaluate the relationship between the expression of platelet-derived growth factor(PDGF) B chain and vascular endothelial growth factor(VEGF) in human glioma and the clinical pathological parameters such as pathological gradation, size, recurrence and prognosis. Methods Applying SABC immunohistochemical technique, we performed examination and statistical research on the expression in 33 cases of human glioma and 5 cases of normal brain tissue. The 33 cases of glioma graded according to WHO 1993 criteria. 13(39.3%) of 33 cases was astrocytoma. 11(33.3%) of 33 cases was anaplastic astrocytoma. 9(27.3%) of 33 cases was glioblastoma multiform. Results In this study, there was obvious differencd in the expression of PDGFBB and VEGF between glioma and normal brain tissue(P<0.01). There was also obvious difference between tumor pathological gradations and the positive expression of PDGFBB and VEGF(P<0.01). Its positive expression increased with elevation of pathological gradation. There was positive correlation between the expression of PDGFBB and VEGF(P<0.05). Conclusion Glioma can secrete PDGFBB and VEGF. There was relationship between the expression of PDGFBB and VEGF and pathological gradation of human glioma, and positive rate increased with elevation of pathological gradation. VEGF might relate with the prognosis of glioma. There was positive correlation between the expression of PDGFBB and VEGF in human glioma.
